The resultant velocity is the velocity of the object with respect to the third frame of reference. Suppose a bird or a plane is moving with respect to the air around it with some velocity V. Suppose the wind is moving with velocity V0 with respect to the ground. Then with respect to the ground the "resultant" velocity of the bird or plane will be V+V0= V' say. Now if we know V' and V0 both with respect to the ground and want to know what is V of the bird or plane with respect to the air, then it is relative velocity V = V'-V0.
For the specific problem you have asked the relative velocity of second man with respect to the first is V2 - V1 and there resultant velocity is V1 + V2 but it does not seem to have any special significance. Here I have considered V1 and V2 as vectors. If they are speeds then the answer for relative speed id V2- V1 if they move in the same direction and V1+V2 if in opposite direction but both are relative velocities, do not go by + or minus sign here.

Answer: We will use nascar as an example since i love nascar. Imagine you are sitting at the track watching two cars race. Each car has a real velocity (car1=150 m.p.h. and car2=200 m.p.h.). Now if you were to jump in car1 and ride around the track at the same speed car2 would now have a relative velocity of 50 m.p.h. Before when you were sitting at the track you could see how fast each car was traveling but when you are in car1 traveling at 150 m.p.h. car2 will appear to be traveling at only 50 m.p.h. (200-150). If the positions were reversed and you were in car2, car1 would appear to be traveling at -50 m.p.h. (falling behind your car) even though you know he is going 150 m.p.h. Relative velocity just depends on where you are measuring the speed.
